摘要
在建立小鼠胚胎干细胞(ES细胞)无饲养层单层培养体系的基础上,使用丁酸钠(NaB)和表皮生长因子(EGF)作为前期诱导分化剂,诱导分化7 d,再用肝细胞生长因子(HGF)和地塞米松(Dex)诱导分化12 d,建立了体外两步法单层诱导ES细胞分化为肝样细胞的方法,并对诱导分化所得的肝样细胞从形态学、基因表达、蛋白分子标记及细胞功能等方面进行了检测,证明了分化所得细胞为肝细胞,肝细胞分化率为41.67%,为肝细胞来源提供了新的方法。
In order to induce mouse embryonic stem(ES)cells to differentiate into hepatocyte-like cells,we employ sodium butyrate and epidermal growth factor as inducer for 7 days in the first phase,followed by the combination of hepatocyte growth factor and dexamethasone as inducer for 12 days in the second phase based on an adherent culture system with two-step induction.We performed the analysis on the morphology,gene expression,protein molecular markers and cell function of ES cells derived hepatocyte-like cells,which showed that the ratio of hepatic differentiation was 41.67%.The ES cell differentiating method provides a new resource for hepatocytes tansplatation.
