摘要
目的:观察兔自体骨髓间充质干细胞(BMSCs)体外诱导分化为心肌样细胞在房室传导阻滞(AVB)模型中的治疗作用。方法:用Percoll密度梯度离心法分离培养兔BMSCs,用5-氮胞苷诱导BMSCs向心肌样细胞分化。在构建AVB模型后第10d实验组于房室结区移植自体心肌样细胞;对照组于房室结区注入培养液。2周后观察实验动物体表心电图,对比移植前后兔AVB的改善情况;冰冻切片观察移植区细胞存活情况。结果:实验组中AVB(-)组的HR、P-P、P-R和R-R间期与AVB(+)组及对照组比有统计学意义(P<0.05);对照组与AVB(+)比无统计学意义。移植区细胞能很好存活。结论:兔自体BMSCs经体外扩增诱导为心肌样细胞后对AVB有治疗作用。
Objective: To observe the therapeutic effect of rabbit autologous bone marrow mesenehymal stem cells(BMSCs), which were induced to cardiomyocyte-like cells in the atrioventricular block(AVB)model. Methods: Rabbit autologous BMSCs were derived and cultured by Percoll continuous density gradient centrifugation, then were induced by 5azacytidine to differentiate into cardiomyocytes-like cells. After ten days of comtructing AVB model, the experimental group was transplanted autologous myocardial-like ceils to the atrioventricular node. The control group was given medium into the atrioventricular node. Two weeks later, by observing surface ECG of the rabbits, improving situation of the mbbits'AVB was compared before and after treatment. The survival cells in transplanted area were also observed by frozen sections. Results: In the experimental group, the heart rate, PP interval, PR interval, RR interval in the AVB(-)group were statistically significant, compared with the control greud and AVB( + )group( P 〈 0.05), there was no statistically significant between control group and AVB(-)group. The transplanted cells survived well. Conclusion: The cardiomyocyte-like cells, which are induced from rabbit autologous BMSCs, have a therapeutic effect in rabbit AVB.
