摘要
目的探讨血小板衍生生长因子及其受体与糖尿病树鼩早期肝纤维化的相关关系。方法将40只树鼩随机分为糖尿病组(n=20),对照组(n=20)。两组动物同步于注射链脲佐菌素(STZ)或生理盐水后8周时进行肝组织活检,标本经血小板衍生生长因子BB(PDGF-BB)及血小板衍生生长因子受体β(PDGFR-β)免疫组织化学染色,在光镜下观察分析PDGF-BB及PDGFR-β在两组动物肝组织的表达。结果①PDGF-BB和PDGFR-β均在两组动物肝血窦壁、肝星形细胞、汇管区动静脉血管内皮细胞、胆管细胞上有阳性表达,糖尿病组PDGF-BB和PDGFR-β的表达程度明显高于对照组。②糖尿病组在不同组织和细胞中PDGF-BB和PDGFR-β表达的阳性数/视野高于对照组(均P<0.01),分别为PDGF-BB肝窦壁86.46±4.85vs 12.78±2.96、肝星形细胞113.75±25.40vs 73.47±12.66、胆管细胞12.35±3.18vs 4.58±1.25、血管内皮细胞25.74±7.13vs 7.29±2.14;PDGFR-β肝窦壁91.52±7.25vs 33.3±3.77、肝星形细胞124.03±23.97vs 85.58±20.64、胆管细胞10.26±2.54vs 4.22±1.60、血管内皮细胞26.83±3.53vs 5.93±1.57。③糖尿病组PDGF-BB/PDGFR-β的表达与胶原纤维的表达的面积密度呈显著正相关(r=0.879,0.850,均P<0.01)。结论 PDGF-BB及PDGFR-β可能参与糖尿病早期肝纤维化的发生、发展。
Objective To study the relationship between platelet-derived growth factor-BB/platelet-derived growth factor receptor β(PDGF-BB/PDGFR-β) and the early stage of liver fibrosis in diabetic tupaiidae. Methods 40 tupaiidaes were divided randomly into diabetic animal model group(n=20) and normal control group(n=20).Liver biopsy was performed after 8 weeks when two groups of animals were injected streptozocin(STZ) or sodium chloride.The expressions of PDGF-BB/PDGFR-β were detected under light microscope by immunohistochemistry of liver specimens. Results ①There were positive expression of PDGF-BB/PDGFR-β on hepatic sinusoid,hepatic stellate cell,bile vessel cell and vascular endothelial cell in both groups of animals,positive expression degree of PDGF-BB/PDGFR-β in diabetic animal model group was higher than that in normal control group obviously.②In diabetic animal model group,positive expression tissue cell numbers per visual field of PDGF-BB/PDGFR-β was higher than that in normal control group,respectively(all P0.01):PDGF-BB on hepatic sinusoid 86.46±4.85 vs 12.78±2.96,hepatic stellate cell 113.75±25.40 vs 73.47±12.66,bile vessel cell 12.35±3.18 vs 4.58±1.25,vascular endothelial cell 25.74±7.13 vs 5.93±1.57;PDGFR-β on hepatic sinusoid 91.52±7.25 vs 33.3±3.77,hepatic stellate cell 124.03±23.97 vs 85.58±20.64,bile vessel cell 10.26±2.54 vs 4.22±1.60,vascular endothelial cell 26.83±3.53 vs 5.93±1.57.③Liver expression of PDGF-BB/PDGFR-β in diabetic animal model group had significantly positive dependablity with collagen area density(r=0.879,0.850,all P0.01). Conclusion PDGF-BB and PDGFR-β may induce and develop liver fibrosis of diabetic tupaiidae.
