Hp26695与宿主细胞作用后烷基过氧化物还原酶研究

Study on the protein of Alkyl Hydroperoxide Reductase after the Interaction of Hp26695 and Host Cells

摘要 目的:鉴定幽门螺杆菌(Hp)与宿主细胞(AGS)作用后的差异蛋白的改变。方法:应用双向电泳分离菌株26695与AGS细胞相互作用前后的蛋白,应用ImageMaster 2Dv3.1软件对蛋白图谱进行比较,对目的蛋白进行基质辅助激光解析及电离飞行时间质谱分析,应用Mascot软件进行蛋白搜库。结果:共监测到5个差异蛋白斑点,经质谱分析鉴定为2种蛋白,分别是热休克蛋白60和烷基过氧化还原酶。结论:对烷基过氧化还原酶深入研究表明,烷基过氧化还原酶可能进入AGS细胞或结合于AGS细胞膜上,这对"幽门螺杆菌可能进入细胞"这一假说给予了有力支持。 Objective: To identify the differential proteins after the interaction of Helicobacter pylori(Hp) and host cells(AGS).Methods: Hp 26695 strain and cell line AGS were used.2-DE and mass spectroscopy were applied to separate protein and identify differential protein.The protein maps were then compared by ImageMaster 2Dv3.1.Mascot software was used for protein database search.Results: Five differential protein spots were found in the gel of 2-DE and identificatied as following 2 proteins:heat shock protein 60(Hsp60)and alkyl hydroperoxide reductase.Conclusion: With the advanced study on the protiens of the alkyl hydroperoxide reductase,results showed that alkyl hydroperoxide reductase could enter the host cells or connect to the membranes of the host cells through the pathogen-cell interactions.This result would give strong support to the hypothsis that the Helicobacter pylori could go into the host cells.
