雷公藤内酯醇对β-淀粉样蛋白诱导的大鼠小胶质细胞IL-1β和PGE_2表达的影响 被引量:3

摘要 目的探讨雷公藤内酯醇对β-淀粉样蛋白(Aβ)1-40诱导的大鼠小胶质细胞白细胞介素-1β(IL-1β)及前列腺素E2(PGE2)表达的影响。方法本实验用Aβ1-40(20μg/ml)诱导体外培养的原代小胶质细胞,建立阿尔茨海默病(AD)细胞模型,并给予不同剂量雷公藤内酯醇(终浓度分别为5、25μg/ml)在不同时程(2 h、12 h、24 h)进行干预,提取细胞培养上清,采用放射免疫学方法检测IL-1β和PGE2的含量。结果加药后12h,模型组IL-1β和PGE2的含量比对照组明显升高(P<0.01);5μg/ml组和25μg/ml组IL-1β的含量比模型组明显减少(P<0.01),5μg/ml组PGE2的含量比模型组明显减少(P<0.01)。结论雷公藤内酯醇对Aβ1-40诱导的小胶质细胞IL-1β和PGE2的表达上调有抑制作用。 Objective To explore the influence of triptolide on the expression of interleukin-1β(IL-1β) and prostaglandin E 2(PGE 2) in rat microglia induced by beta-amyloid protein(Aβ).Methods Microglia was induced by Aβ1-40(20 μg/ml) to establish the AD cellar model,then treated with triptolide(final concentration respectively was 5,25 μg/ml) for 2,12,24 h.The supernatants were collected for detecting the contents of IL-1β and PGE 2 by radioimmunoassay.Results After 12 h of cultivation,the contents of IL-1β and PGE 2 in model group were obviously increased compared with those of control group(P 0.01).The contents of IL-1β in 5 and 25 μg/ml groups were obviously decreased compared with those of model group(P 0.01).The content of PGE 2 in 5 μg/ml group was obviously decreased compared with that of model group(P 0.01).Conclusions Triptolide can suppress the up-expression of IL-1β and PGE 2 in rat microglia induced by Aβ1-40.
