α苦瓜素对大鼠长期毒性实验的血液学指标分析

摘要 目的探讨长期毒性实验中α苦瓜素(α-MMC)对大鼠的血液学影响。方法将SD大鼠随机分为生理盐水对照组、α苦瓜素各剂量组(6.25,2.08,0.70mg/kg),隔日经尾静脉注射给药一次,连续28天,停药后恢复14天,分别在给药期末和恢复期末进行血液常规检查、流式细胞分析和骨髓象检查。结果连续给予大鼠α苦瓜素28天后,随着剂量的增加,WBC、LYM、RBC、HGB逐渐减少,NEU和PLT则逐渐增多。经过14天的恢复期,各血液学指标的异常变化有所改善。结论长期给予α-MMC会引起大鼠以血细胞数量变化为主的血液学毒性变化。 Objective To explore hematologic effects of α-MMC on rats in long-term toxicity.Methods SD rats were randomized into NS control group and α-MMC groups.α-MMC of 6.25 mg/kg,2.08 mg/ kg and 0.70 mg/kg and normal saline in 0.2ml were administered to rats respectively via caudal vein every other day for consecutive 28 days and then recovery for 14 days.On day 28 after initiation of administration and on day 14 after withdrawal,samples were collected for blood routine examination,flow cytometry and bone marrow smear test.Results On day 28 after initiation of administration,with increase of dose,WBC,NEU,RBC,HGB were decreased gradually,but NEU and PLT were increased.After recovery for 14 days,the abnormal change of hematologic parameters had some improved.Conclusion α-MMC can lead to hematologic toxic changes especially the number of blood cells if SD rats were administered by long term.
