摘要
目的研究p27在面神经纤维压榨伤后大鼠面神经核中的表达变化及其作用。方法制做成年大鼠面神经压榨伤模型,观察大鼠行为变化,利用免疫组织化学技术观察大脑面神经核运动神经元中p27蛋白表达的时程变化。结果面神经纤维压榨伤后压榨侧触须垂下,拂动消失,眼睑不能闭合。1周后面神经功能开始恢复,4周后完全恢复正常。检测面神经压榨侧6h、12 h、1周、2周、3周、4周等时间点p27含量均高于正常侧,差异有统计学意义(P<0.05),而在1 d、2 d、3 d时压榨侧与健侧相比无统计学差异﹙P>0.05﹚。结论面神经压榨伤后早期p27升高可能与机体的应激反应有关,晚期p27升高的机制可能是与其参与损伤神经元的DNA修复有关。
Objective To investigate temporal expression of p27 in rat facial nucleus after facial nerve crush injury. Methods The adult rat model of facial nerve crush injury was established and rat behavior after the surgery was observed. The temporal variation of p27 expression in the motor neurons of facial nucleus was examined by immunohistochemistry. Results After the facial nerve crush injury,the whiskers of the injured side bent down, the whisker movement disappeared and the eyelid could not close. After 1 week, the motor function of facial nerve motor began to recover and reached normal level after 4 weeks. The p27 expression in the injured side was significantly higher than that in the normal side at 6 h, 12 h, 1 week,2 weeks,3 weeks and 4 weeks (P 〈 0.05),while there was no significant difference between the injured side and the normal side at 1 d, 2 d and 3 d (P 〉 0.05 ). Conclusion The early increase of p27 expression might be related to stress reaction after facial nerve crush injury. The late upregulation of p27 expression might be related to its involvement in the DNA repair of the damaged neurons.
