绵羊肺腺瘤病毒人工感染羊重要病理生理指标的检测

摘要 进一步探讨人工感染绵羊肺腺瘤病毒的羊对淋巴细胞亚群、淋巴细胞凋亡、血气指标、肿瘤标志物的影响。对已人工感染JSRV 300d的病羊,进行临床指标测定,每隔30d用流式细胞仪检测外周血中淋巴细胞及其亚群的动态变化以及淋巴细胞凋亡情况。同时,对血液做血气分析、血清酶和肿瘤标志物的检测。结果:SPA攻毒组感染JS-RV-NM株300d后,外周血中CD4+细胞水平较低;CD8+呈现出高低波动的动态变化过程。对淋巴细胞凋亡检测的结果显示,在感染第330d,凋亡细胞都保持较高水平;而感染第360d和390d,凋亡细胞水平都降低。血气分析结果显示pCO2、pO2、Hct、BE(ecf)、BE(B)、ctCO2、AnGap、O2SAT、O2CT、ctHb(est)、Cl-十一项在不同时期与对照组存在显著性差异。肿瘤标志物CEA和AFP检测结果均在正常范围内,但攻毒组均高于对照组。结论:病羊在感染绵羊肺腺瘤病毒300d后,血液中CD4+T淋巴细胞和CD8+T淋巴细胞有变化,这是SPA病羊在疾病过程中细胞免疫和淋巴细胞受损的重要表现之一;外周血淋巴细胞会呈现不同程度的凋亡,绵羊肺腺瘤病毒诱导机体免疫细胞凋亡是一种重要的临床表现;本实验结果证明血气分析的检测在诊断绵羊肺腺瘤病时有一定价值,说明实验羊发生呼吸功能障碍。攻毒组的肿瘤标志物癌胚抗原(CEA)和甲胎蛋白(AFP)均高于对照组,但其病理生理意义和诊断价值尚待进一步研究。 OBJECTIVE : To further discuss the effects on lymphocytes subsets, lymphocytes apoptosis. Blood - gas. tumor markers of sheep which artificially infected with Sheep Pulmonary Adenomatosis Virus. METHODS: To detect the clinical indicator for the sheep who have been infected for 300 days. At 330, 360 and 390d , used the flow cytomytor to examine lymphocytes apoptosis number, CD4 * T and CD8 * T lymphocytes, Analysis of blood flowas, Serum enzymes and to detect tumor markers. RESULTS : After affected 330d, the level of CD4 +cells was low; the level of CD8 * sometimes was low and sometimes was high. At 330 d post - infection, CD4 + T, CD8 + T lymphocytes apoptosis were higher than that of control, and decreased at 360 d and 390 d. pCO2 , pO2 ,Hct, BE (eef) , BE (B) ,ctCO2 , AnGap, O2SAT, O2CT, ctHb (est), C1- showed significant difference between the experimental group and the control group. CA125 and CEA were normal,but the value of infected sheep were higher. CONCLUSIONS: Sheep have been infected for 300 days, the changes of CD4 + T, CD8 +T lymphocytes in peripheral blood of sheep artificially infected with sheep pulmonary adenomatosis virus may be the result of cell immunological reaction and the lymphocytes injury. Periphery lymphocyte apoptosis in sheep artificially infected with sheep pulmonary adenomatosis virus was found,lymphocyte apoptosis may be one of the most important clinical symptoms. The detection of blood - gas of sheep artificially infected with sheep pulmonary adenomatosis virus was helpful for diagnosis of SPA. The worth of detecting CEA and AFP need more research.
