摘要
通过比较大米中角鲨烯的不同提取方法,选用优化后的索氏提取/气相色谱-串联质谱(GC-MS/MS)法对不同产地大米中的角鲨烯进行定性及定量分析。结果表明,该方法在1~50 mg/L质量浓度范围内线性良好,相关系数为0.998 7;检出限为0.1 mg/kg;在5、20 mg/kg 2个加标水平下的回收率可达96%~104%;相对标准偏差(RSD)不大于3.6%。对不同产地大米中角鲨烯的分析表明,该方法的灵敏度高、重复性好,可作为区分大米不同种属、产地和品质的有效方法。
Different extraction methods of bioactive component squalene in rices were studied and a new method was developed for the determination of squalene in rices. The squalene in rices from different areas were extracted by Soxhlet extract method, and analyzed by gas chromatography -tandem mass spectrometry(GC -MS/MS). The results showed that the calibration curve was linear over the concentration of squalene in the range of 1 - 50 mg/L with a correlation coefficient of 0. 998 7. The detection limit(LOD) was 0. 1 mg/kg. The recoveries of squalene at two spiked levels of 5 mg/kg and 20 mg/kg were in the range of 96% -104% with relative standard deviations(RSDs) no more than 3.6% . The method was sensitive and reproducible, and could be used to distinguish effectively rices with different species, origins and qualities.
