调脂颗粒对小鼠高脂血症的影响 被引量:9

摘要 目的观察调脂颗粒(泽泻、姜黄、蒲黄和莱菔子)对小鼠实验性高脂血症的影响。方法采用脂肪乳剂灌胃的方法建立高脂血症小鼠模型,将50只雄性昆明小鼠随机分为正常组、高脂模型组、阳性药力平之组和调脂颗粒小、大剂量组,每组10只,给药5周后分别观察调脂颗粒对高脂血症小鼠血清总胆固醇(TC)、甘油三酯(TG)、游离脂肪酸(FFA)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、谷丙转氨酶(ALT)、谷草转氨酶(AST)以及体质量、肝质量系数、肝组织内TC、TG、FFA水平的影响;光镜观察小鼠肝组织脂肪变性程度;免疫组织化学方法观察小鼠肝组织内过氧化物酶体增殖物激活受体γ(PPARγ)和肝型脂肪酸结合蛋白(L-FABP)表达的变化。结果高脂饮食小鼠灌服1.25g/kg2、.5 g/kg调脂颗粒5周后,血清TC、TG、FFA、LDL-C、ALT和AST以及体质量、肝质量系数、肝组织内TC、TG、FFA水平均降低(P<0.05,P<0.01),血清HDL-C水平明显升高(P<0.01)。光镜检查结果表明,给予调脂颗粒的小鼠肝组织脂质变性程度明显减轻(P<0.01)。免疫组织化学结果显示,调脂颗粒组小鼠的肝脏内PPARγ和L-FABP蛋白表达水平升高(P<0.05,P<0.01)。结论调脂颗粒可抑制小鼠高脂血症的形成,其机制可能与上调肝组织中PPARγ及L-FABP蛋白的表达,加速对脂质的转运和代谢有关。此外,调脂颗粒还具有一定的肝脏保护作用。 AIM To investigate the inhibiting effect of Tiaozhi Granule(Alismatis Rhizome,Curcumae longae Rhizome,Typhae pollen,Raphani Semen) on the experimental hyperlipidemia in mice.METHODS A mouse model for hyperlipidemia was induced by feeding fatty milk.Fifty Kunming mice were randomly divided into 5 equal groups: normal control group,model control group,Lipanthyl group,Tiaozhi Granule low dose group,and high dose group.After five weeks,the serum levels of TC,TG,FFA,HDL-C,LDL-C,ALT and AST,the body weight and the coefficient of hepatic weight as well as the hepatic TG,TC,FFA contents were inspected,respectively.Hepatic fatty degeneration of mice livers was observed under light microscopy.The protein expressions of PPARγ and L-FABP in hepatic tissue were detected by immunohistochemistry method.RESULTS After the administration of 1.25 and 2.5 g/kg Tiaozhi Granule,the serum levels of TC,TG,FFA,LDL-C,ALT,AST,the body weight and the coefficient of hepatic weight as well as the hepatic TC,TG,FFA contents decreased compared with the hyperlipidemic model group(P〈0.05 or P〈0.01),while the serum HDL-C level increased obviously(P〈0.01).The histological evaluation of mice livers under light microscopy showed that the degree of fatty degeneration in Tiaozhi Granule treated groups was dramatically improved(P〈0.01).The immunohistochemistry results indicated that the protein expressions of hepatic PPARγ and L-FABP were up-regulated in the hyperlipidemic mice after the treatment of Tiaozhi Granule(P〈0.05 or P〈0.01).CONCLUSION Tiaozhi Granule might inhibit the hyperlipidemic formation in mice,and the mechanism might be related to the acceleration of lipid transfer and the metabolism via the up-regulation of L-FABP and PPARγ protein expressions.Moreover the drug possesses a protective effect on liver.
