摘要
以野牡丹科植物幼叶为材料,对影响野牡丹科植物DNA提取质量的主要因子进行研究,以期建立适宜野牡丹科植物DNA提取的优化反应体系。结果表明,当提取液中CTAB浓度为4%,沉淀时加入0.6倍体积预冷异丙醇、1/2倍体积5 mol/L NaCl、2倍体积无水乙醇时,所提取的DNA纯度较高,电泳条带清晰。且在此条件下,能提出野牡丹科7种植物的DNA。
Young leaves of the Melastomaceae plants were used as experimental materials, the main factors affecting DNA extract quality from the Melastomaceae plants were studied in order to establish an optimal reaction system of DNA extraction. The results indicated that when the CTAB concentration of extract was 4%, and added 0.6 volume of ice-cold isopropanol, 1/2 volume of 5moFL NaC1 and 2 volumes of anhydrous ethanol when it precipitating, the purity of extracted DNA were higher, and the electrophoretic bands were clear. And in this condition, the DNA of 7 species plants of Melastomaceae could be extracted.
