摘要
目的:探讨精神分裂症患者外周血淋巴细胞多巴胺D2受体(DRD2)与转运体(DAT)的mRNA表达水平及其与临床症状的关系。方法:研究对象为2009年12月~2010年10月间在无锡市精神卫生中心住院的首发精神分裂症患者25例及同期在该医院体检的健康成人28例。所有被试者空腹抽取肘静脉血2 ml,采用病例对照研究,RT-PCR技术检测精神分裂症患者及正常人外周血淋巴细胞DRD2、DAT的mRNA表达情况。用阳性阴性症状评定量表(PANSS)评定精神分裂症患者的临床症状和严重程度。结果:精神分裂症患者外周血淋巴细胞DRD2、DAT表达较对照组显著增高(P<0.05),精神分裂症患者外周血淋巴细胞DRD2表达与阳性症状分呈正相关(r=0.456,P=0.022),DAT mRNA表达水平统计结果无统计学意义(P>0.05)。结论:精神分裂症患者外周血淋巴细胞DRD2 mRNA表达水平显著增强,其表达水平可以作为精神分裂症早期诊断和筛查的一个外周标记。
Objective:To investigate the protein level of dopamine receptor D2(DRD2) and dopamine transporter(DAT) in peripheral blood lymphocytes(PBL) in patients with schizophrenia and clinical symptom demonstration.Methods:By case-controls study,we included 25 schizophrenics experiencing their first episode and seeking for hospitalization in Wuxi Mental Health Center between Dec.2009 and Sept.2010 and another 28 healthy adults who underwent physical examination in the corresponding period.All subjects were subjected to fasting venous blood taking of 2 ml for detecting mRNA expression of DRD2 and DAT by reverse transcription polymerase chain reaction(RT-PCR).Positive and Negative Symptom Scale(PANSS) was used to estimate the clinical symptoms and severity for patients with schizophrenia.Results:The levels of DRD2 and DAT were significantly higher in PBL of schizophrenic as compared with the controls(P0.05),and DRD2 expression was positively associated with positive symptom in PANSS(r=0.536,P=0.006;r=0.604,P=0.001).Yet,DAT mRNA levels revealed no statistical difference(P0.05).Conclusion:Elevated DRD2 mRNA expression was found in PBL in patients with schizophrenia,suggesting that the protein level can be used as a peripheral marker for early diagnosis and screening of this disorder.
