摘要
目的检测原发性肝细胞癌RASSF1基因启动子甲基化状态并探索其作为新的肿瘤生物标志物在肝细胞癌诊断中的意义。方法应用MSP和RT-PCR分别检测60例原发性肝细胞癌组织样本和癌旁组织RASSF1基因启动子甲基化状态和mRNA表达。结果 60例PHCC组织样本中有51例存在RASSF1基因启动子甲基化状态(85.0%),均检测不到mRNA表达,癌旁组织甲基化阳性率为5%,mRNA表达正常。RASSF1基因启动子甲基化与肿瘤病理分期和直径大小呈正相关关系,而与患者性别、年龄和血清AFP水平无关。结论 RASSF1基因启动子甲基化在PHCC的发病机制中起到重要作用,可以作为新的肿瘤生物标志物为PHCC诊断和评估提供重要分子线索。
【Objective】 To detect the methylation status of RASSF1 gene promoter in primary hepatocellular carcinoma(PHCC) and explore its biomarker role in the diagnosis of PHCC.【Methods】 MSP and RT-PCR were conducted to detect RASSF1 gene promoter's methylation status and its mRNA expression in PHCC and paracancerous tissues respectively.【Results】 51 in 60 PHCC tissues displayed positive methylation status(85.0%) with no mRNA expression,while it was 5% in paracancerous tissues with normal mRNA expression.RASSF1 gene promoter methylation positively correlated with clinical tumor staging and its size,while had nothing to with patients' gender,age and serum AFP levels.【Conclusions】 RASSF1 gene promoter hypermethylation may play a crucial role in the pathogenesis of PHCC,it could be served as a tumor biomarker and provide important molecular clues in the diagnosis and evaluation of PHCC.
