大黄素对急性肝衰竭大鼠肝功能的影响 被引量:4

摘要 目的探讨大黄素对急性肝衰竭(ALF)大鼠肝功能的影响。方法 SD大鼠随机分三组:健康对照组、模型(ALF)组、大黄素组。D-氨基半乳糖(D-Gal)诱导大鼠急性肝功能衰竭模型,大黄素组于造模后12h开始腹腔注射大黄素10mg/kg,每天2次。模型组及大黄素组分别在造模后24、72和120、168h,随机各取6只,留取门静脉血及肝组织。结果造模后24h,大鼠血清ALT、AST值明显升高,分别为(537.3±133.7)U/L和(797.2±159.3)U/L,显著高于正常组的(34.5±4.2)U/L和(81.6±5.1)U/L,差异有统计学意义(t=-5.579、-4.825,P<0.01),以72h最为显著。造模后24h,大黄素组与模型组血清ALT、AST差异无统计学意义,造模后72h,大黄素组血清ALT、AST分别为(116.8±56.2)U/L和(165.5±78.7)U/L,显著低于模型组的(728.0±137.4)U/L和(1416.7±254.7)U/L,差异有统计学意义(t=-6.735、-6.544,P<0.05)。至120h和168h,两组血清ALT、AST均持续下降。结论大黄素具有改善急性肝衰竭大鼠肝功能的作用,具体机制有待进一步研究。 Objective To study the effect of emodin on liver function in rat model of acute liver failure ( ALF ). Methods SD rats were divided into three groups, there were six rats in normal group and twenty four in model group of ALF and emodin group. The model of ALF was induced in rats by D-galactosamine ( D-Gal ) successfully. The sera and hepatic tissue samples were collected at 24,72, 120 and 168h after D-Gal injection. The sera were used for detecting ALT and AST. The pathologic variation in hepatic tissue samples were detected by H-E stain. Results The levels of alanine animotransferase ( ALT ) and aspartate animotransferase ( AST ) at 24h were ( 537.3 ± 133.7 ) U/L and ( 797.2 ± 159.3 ) U/L, respectively, which were both significantly higher than those in normal group, showing great difference between 24h group and normal group[ ( 34.5 ± 4.2 )U/L and ( 81.6 ± 5.1 )U/L] ( t=-5.579,-4.825 ,P 〈 0.01 ), peaked at 72h after establishing the model of ALF. The levels of ALT and AST in model group at 24h had no significantly difference than those in emodin group, and at 72h, the levels of ALT and AST in emodin group[ ( 116.8 ± 56.2 ) U/L and ( 165.5± 78.7 ) U/L] were lower significantly than those in model group[ ( 728.0 ± 137.4 ) U/L and ( 1416.7 ± 254.7 ) U/L] ( t=-6.735, -6.544, P 〈 0.05 ). Conclusion Emodin can obviously decline the plasm levels of ALT, AST in rat model of acute liver failure, the mechanisms for this effect still needs more studies.
