摘要
目的探讨MTT法在原代肺癌细胞药物敏感性试验研究中的应用价值及NSCLC对5种(长春瑞滨、依托泊苷、长春新碱、吉西他滨、多西他赛)临床常用化疗药物的敏感性。希望能通过本课题的研究为肺癌个体化化疗敏感药物的选择提供参考。方法 30例病例均取自手术新鲜组织,采取体外原代培养,MTT药敏试验,检测5种(长春瑞滨、依托泊苷、长春新碱、吉西他滨、多西他赛)临床常用化疗药物的敏感性,选取临床最佳化疗方案。结果 30例NSCLC体外原代培养均成功。肺癌细胞对5种化疗药均敏感,其顺序依次为多西他赛=吉西他滨>长春瑞滨=依托泊苷=长春新碱。结论本课题之肺癌细胞原代培养的方法可获得药敏实验所需的肺癌细胞。MTT法在原代肺癌细胞药物敏感性试验研究中具有灵敏度高、重复性好、操作简便、经济、快速、易自动化、无放射性污染等优点,值得推广。30例非小细胞肺癌细胞对多西他赛和吉西他滨最敏感,可作为临床治疗NSCLC首选之一,该实验方法可作为NSCLC个体化治疗的选择之一。
Objective To stuty the original generation cultivation of tumor cells of 30 patients of this article diagnosed with NSCLC and individual susceptibility test research, that was aimed to explore the application value of MTT method for the origenal lung cancer cells drug sensitivity test study and NSCLC was sensitive to five (TAX, GEM, NVB, VP-16, VCR) drugs that used clinically chemotherapy. Hopefully through this topic research we can reference for individualized sensitive chemotherapy of lung cancer. Methods 30 cases were from the fresh tissue, adopting the original generation Cultivation and MTT susceptibility test, testing sensitivity of five ( TAX, GEM, NVB, VP- 16,VCR) chemotherapeutic drugs, selecting clinical best chemotherapy regimen. Results 30 ca- ses of NSCLC in vitro original generation cultivation all success, Lung cancer ceils were sensitive to 5 kinds of chemotherapeutic drugs, the sequence was TAX = GEM 〉 NVB = VP-16 = VCR. Conclu- sion The original generation cultivation method of the lung cancer cells of this topic can raise the success rate. MTT method in lung cancer cells drug sensitivity test study compared with other meth- od had high sensitivity, repeatability, operation simple, economic, quick, easy automation, no radioactivity pollution etc ,was worthy to be popularized, the most sensitive drug was TAX and GEM in 30 none small cell lung cancer cells, that can be used as one of the clinical cureing NSCLC pre-ferred chemotherapy method, this can be used as one of individualized sensitive chemotherapy of NSCLC.
