Effect of ray cartilage extract on human mammary carcinoma cell line MCF-7

摘要 目的研究魟鱼骨素(RCE)对人乳腺癌细胞系MCF-7细胞生长周期的影响。方法以不同浓度RCE处理MCF-7细胞不同时间,采用MTT法检测各组MCF-7细胞增殖情况,流式细胞仪检测药物作用前后对细胞周期的变化,Western blot检测药物作用前后细胞周期素D1及p21蛋白表达。结果 RCE对MCF-7细胞增殖的抑制呈浓度依赖性和时间依赖性(P<0.05),且存在最大抑制浓度10μmol/L。随着RCE作用时间延长,S期细胞百分比明显增加,而G2/M期细胞比例明显减少(P<0.05),且细胞凋亡率随药物作用时间的延长明显升高(P<0.05)。随着药物作用时间延长,MCF-7细胞中的细胞周期素D1表达降低,p21蛋白表达逐渐增高(P<0.05)。结论 RCE抑制MCF-7细胞增殖,阻滞细胞从S期向G2期转化;其机制可能与调控细胞周期蛋白表达有关。RCE可能成为未来治疗乳腺癌药物之一。 Objective To investigate the effect of ray cartilage extract(RCE) on human mammary carcinoma cell line MCF-7.Methods The MCF-7 cells were treated with different concentrations of RCE for different durations.MTT assay was used to evaluate the proliferation of the cells.The effect of RCE on the cell cycle was detected by flow cytometry.The expressions of cyclin D1 and p21 in MCF-7 cells were determined by Western blot.Results The proliferation of MCF-7 cells was inhibited in the concentration-and time-dependent manner(P0.05) and the maximal inhibition was seen at a concentration of RCE 10 μmol/L.The percentage of S phase cells was increased and that of G_2/M phase cells decreased,and MCF-7 apoptosis increased as RCE-acted duration prolonged in MCF-7 cells(P0.05).The expression of cyclin D1 decreased and that of p21 increased with prolongation of the RCE-acted duration(P0.05).Conclusion RCE inhibits MCF-7 cell proliferation and its transformation from S phase to G_2 phase,which may be related to the inhibition of the cyclin D1 expression and the increase of the cyclin p21 expression.RCE could be a potential drug to treat breast cancer.
