摘要
采用不同的破碎方法对同一株醋酸菌菌体进行破碎,结果表明,采用溶菌酶-超声波联合破碎法,破碎效果更佳,ADH酶活力也更高。通过单因素和正交试验确定溶菌酶-超声波联合破碎法的最佳破碎条件:溶菌酶用量为0.04mg/mL,酶解时间为20min,酶解温度为30℃,超声波累计破碎时间为2.5min。
The disruption method cooperated with lysozyme reaction and ultrasonic together is better than disruption lysozyme reaction or ultrasonic alone on the disruption of an acetic acid bacteria to extract ADH.The optimize parameters of disruption method with lysozyme reaction and ultrasonic were obtained by single factor experiment and orthogonal experiment.The results show that the reaction dosage of lysozyme was 0.04 mg/mL,the reaction time was 20 min,the reaction temperature was 30 ℃,the disruption time with ultrasonic was 2.5 min.
