摘要
目的检测Ⅰ型胶原(Col-Ⅰ)/聚乙烯醇(polyvinyl alcohol,PVA)共聚纺丝的生物相容性。方法使用小鼠成纤维细胞(L-929),分为100%浓度Col-Ⅰ/PVA共聚纺丝浸提液、50%浓度Col-Ⅰ/PVA共聚纺丝浸提液、空白对照、阳性对照4个组,MTT法测定各组细胞在1、2、3、4、5、6、7 d的光密度值,计算细胞相对增殖率(relative growth rate,RGR),判定细胞毒性的级别;直接接触共培养L-929细胞,倒置显微镜和扫描电镜观察细胞在Col-Ⅰ/PVA共聚纺丝上的生长情况;将Col-Ⅰ/PVA共聚纺丝和聚对苯二甲酸乙二醇酯(PET)纤维分别植入昆明小鼠皮下,于术后1、3、9周取标本大体观察包膜情况,HE、Mallory三色法进行组织学观察。结果 MTT法:2个实验组细胞在不同时间点的相对增殖率均为90%~123.61%,Col-Ⅰ/PVA共聚纺丝的细胞毒性评级为0~Ⅰ级;直接接触法:细胞与Col-Ⅰ/PVA共聚纺丝直接接触共培养,生长良好、形态正常,并附着生长。皮下植入的Col-Ⅰ/PVA共聚纺丝和PET纤维在1、3、9周均有包膜形成,且随时间的延长包膜透明度增高,包膜周围炎细胞主要为中性粒细胞、巨噬细胞、淋巴细胞,随时间的延长炎细胞数量明显减少。结论 Col-Ⅰ/PVA共聚纺丝的生物相容性良好,且细胞毒性和组织相容性均在临床应用的允许范围内。
Objective To test the biocompatibility of type I collagen and polyvinyl alcohol(Col-Ⅰ/PVA) co-polymerizing filature.Methods Mouse fibroblast L-929 cells were cultured in the medium contain-ing 100% or 50% consistency Col-Ⅰ/PVA co-polymerizing filature leaching liquor.Those cells cultured in normal medium or in the medium contain 0.5% phenol solution served as blank control and positive control,respectively.MTT assay was used to test absorbance value of the cells on the 1st,2nd,3rd,4th,5th,6th,and 7th day.Thus,relative growth rate(RGR) of the cells was calculated,and their cytotoxicity was graded.Direct contact method was used to culture L-929 cells directly on Col-Ⅰ/PVA co-polymerizing filature.The cell growth was observed by inverted microscopy and scanning electron microscopy.Col-Ⅰ/PVA co-polymerizing filature and polyethylene terephthalate(PET) fiber were respectively implanted subcutaneously into KM mice.Specimens were harvested at 1st,3rd and 9th postoperative week for gross observation,and histological changes through HE and Mallory triple staining.Results Relative growth rate of the cells in 100% and 50% consistency Col-Ⅰ/PVA co-polymerizing filature leaching liquor varied from 90% to 123.61% at different time points.Cytotoxicity of Col-Ⅰ/PVA co-polymerizing filature was graded at 0 to Ⅰ.The cells were cultured together with Col-Ⅰ/PVA co-polymerizing filature by means of direct contact.The growth of these cells was in good condition,with normal shapes and attaching to Col-Ⅰ/PVA co-polymerizing filature.Capsules were formed around both subcutaneously implanted Col-Ⅰ/PVA co-polymerizing filature and PET fiber at 1st,3rd and 9th week,with their transparency increasing with the elapse of time.The inflammatory cells around the capsule were mainly neutrophilic granulocytes,macrophages and lymphocytes,whose amounts were obviously decreased with the extension of time.Conclusion Col-Ⅰ/PVA co-polymerizing filature has good biocompatibility.Its cytotoxicity and histocompatibility are within the permission of clinical application.
