摘要 背景:肿瘤干细胞是否存在于所有肿瘤中仍有争议。目的:从人胃癌细胞中分离并鉴定类肿瘤干细胞,对胃癌的发生机制进行探讨。方法:原代培养胃癌细胞,并通过流式细胞术检测CD44和CD29等细胞因子的阳性表达率,分选阳性细胞进行成脂诱导。结果与结论:胃癌细胞中CD44、CD29的阳性百分率分别为5.67%和5.53%,CD44和CD29阳性细胞具有成脂能力,证明胃癌肿瘤细胞中存在具有分化能力的类肿瘤干细胞。 BACKGROUND:Whether cancer stem cells are present in all tumors is still controversial.OBJECTIVE:To isolate and identify cancer stem cells from human gastric cancer cells,and to explore the mechanism of gastric cancer.METHODS:Gastric cancer cells were primarily cultured and CD44 and CD29 positive expression rates were detected by flow cytometry to sort positive cells for adipogenic induction.RESULTS AND CONCLUSION:CD44 and CD29 positive percentages in gastric cancer cells were 5.67% and 5.53%,respectively,indicating that CD44 and CD29 positive cells had an adipogenic capacity.It is proved that there are tumor stem cells with differentiation capacity in gastric cancer cells.
