摘要
目的:建立同时测定何首乌中大黄素、大黄酸、大黄酚、大黄素甲醚、芦荟大黄素5种游离蒽醌类成分含量的方法。方法:色谱柱为Agilent Zorbax SB-C18(5μm,150mm×4.6mm),流动相为0.02%磷酸水溶液-乙腈,梯度洗脱,流速为1 mLmin-1,检测波长254nm。结果:5条标准曲线在检测范围内均呈良好线性(r≥0.9999);5个待测成分的检测限均低于0.18ng,定量限均低于0.57ng;日内和日间精密度的RSD均小于3.1%,加样回收率为91.0%~103.1%。结论:本文所建立的何首乌中5个指标成分同时定量的方法准确、可靠,可作为何首乌药材质量控制的参考方法。
Objective:To establish the method for simultaneous determination of 5 free anthraquinione.derivatives from fleeceflower Root. including emodin, rhein, chrysophanol, physcion, aloe-emodin,s ennoside A and sennoside B. Methods:The analysis was achieved with an Agilent Zorbax SB-C18 analytical eoinmn (150 mm× 4.6 mm,5μm) by gradient eintion of 0.02%(v/v) phosphoric acid in water and acetonitrite.The flow rate was 1 mLmin-1,and detection wavelength was set at 254 nm. Results: All the calibration curves showed good linearity (r≥0.9999) in a relatively wide concentration range. The LOD and LOQ were lower than 0.18 ng and 0.57 ng on column, respectively. The RSD for intra- and inter-day of 5 analytes was less than 3.1%, and the recoveries were 91.0%-103.1%.The validated method was successfully applied for the analysis of 5 free anthraquinone in fleeceflower Root. Conclusion: The developed HPLC assay is accurate and reliable, and can be readily utilized as a quality control method for fleeeeflower Root.
