摘要
目的探讨法舒地尔对大鼠缺血性脑水肿脑组织中AQP9基因表达和蛋白合成的影响。方法健康SD大鼠144只,随机分为:对照组、手术组、法舒地尔组,建立大脑中动脉闭塞(MCAQ)模型,分别在6 h、24 h、72 h和5 d 4个时间点将大鼠麻醉后断头取脑,采用干湿比重法测脑组织含水量,行HE染色,并用R T-PCR、Western Blot检测脑组织中AQP4基因表达和蛋白合成的变化。结果与对照组相比,手术组、法舒地尔组均从6 h开始出现脑水肿,72 h时达到高峰,且法舒地尔组脑水肿低于手术组;同样AQP9及其mRNA的表达在手术后6 h后开始升高,72 h后达到高峰,且手术组较对照组相和法舒地尔组明显增高(P<0.05)。结论 AQP9与脑水肿形成有关,法舒地尔可以降低脑组织内AQP9的基因表达和蛋白合成从而减轻脑水肿。
【Objective】To study effects of fasudil on the expression of AQP9 mRNA and AQP9 after ischemic brain edema in rat.【Methods】144 normal SD rats were randomized to controlled group,surgery group and fasudil group.AQP9 mRNA and AQP9 expression were determined by RT-PCR and western blot after middle cerebral artery occlusion(MCAO) at 6h,24h,72h,5d.At each time point,the water contents in the brain tissues were measured as wet-to-dry weight ratio.The pathology was checked at each time too.【Result】Compared with the control group,cerebral edema appeared in surgery group and fasudil group 6h after operations,it reached peak at 72h and cerebral edema of Fasudil group were lower than surgery group.The AQP9 mRNA and AQP9 expression of surgery group were obviously higher than those in the controlled group and fasudil group(P 0.05),and they increased at 6h,reached peak at 72 h.【Conclusion】AQP9 may be involved in the formation of brain edema,Fasudil can decrease the expression of AQP9 mRNA andAQP9 and relieve brain edema.
