摘要
目的:探讨经复方苦参注射液处理肝癌细胞(HepG2)后survivin mRNA表达。方法:采用MTT比色法测定肝癌细胞对氧化苦参碱的敏感性。RT-PCR检测survivin mRNA的表达。Western-blot检测survivin蛋白表达的变化。结果:复方苦参注射液处理后的第1天,3组肝癌细胞的survivin mRNA表达水平均降低;其中0.1 g.mL-1复方苦参注射液组下降了5%,0.2 g.mL-1复方苦参注射液组下降8%,0.3 g.mL-1组下降了28%。第3天,0.1 g.mL-1组和0.2 g.mL-1组HepG2细胞的survivin mRNA表达与第1天比较,分别下降10%和36%,0.3 g.mL-1组仍持续降低,为第1天的66%。0.1 g.mL-1组和0.2 g.mL-1组作用3 d后的HepG2细胞中survivin蛋白含量分别下降了3%和15%,0.3 g.mL-1组则下降到了55%。结论:复方苦参通过调控survivin mRNA在肝癌细胞的表达,可能是其抗肿瘤作用的机制。
OBJECTIVE To explore the relationship between survivin and chemoresistance, and to study the alteration of survivin mRNA and protein expression after treatment with compound Radix Sophorae Flavescentis injection. METHODS RTPCR and Western blot were used to analyze the changes in expression levels of survivin mRNA and protein. HepG2 cell was examined with MTT assay. RESULTS The level of survivin mRNA was descent when HepG2 cell treated with three kinds of concentration of compound Radix Sophorae Flavescentis injection for one day. The survivin mRNA treated with 0. 1 go mL 1 was descent 5 %, descent 8 % for 0. 2 g. mL- 1 compound Radix Sophorae Flavescentis injection, and descent 28 % for 0. 3 g·mL^-1 In the third day, the survivin level treated with 0. 1 gomL-1 was descent 10 %, 0. 2 g.mL- 1 descent 36%, 0. 3 g.mL-1 compound Radix Sophorae Flavescentis injection descent 66%//00. Similarly, survivin protein level was descent 3% for treated with 0.1g.mL^-1 after three days, descent 15%for 0. 2 g.mL-1, descent 55% for 0. 3 g.mL 1. CONCLUSION Compound Radix Sophorae Flavescentis injection which by regulating survivin mRNA expression in hepatoma cells enhances their sensitivity to chemotherapy of liver cancer cells; this may be the mechanism of its antitumor effect.
