天麻与钩藤配伍前后天麻素在SHR大鼠肝脏和肾脏的分布与归经探讨 被引量:4

摘要 目的:建立SHR大鼠肝、肾组织中天麻素含量的测定方法,并通过天麻与钩藤配伍前后效应成分天麻素的组织分布差异探讨归经。方法:采用高效液相色谱法测定SHR大鼠肝、肾组织中天麻素的含量。色谱柱为Phenomenence Luna C18(250mm×4.6mm,5μm),流动相为乙腈-1%磷酸(3∶97),流速为1.0mL·min-1,检测波长为220nm,柱温为30℃,进样量为10μL。结果:天麻素的检测浓度在0.3125~200μg·mL-1范围内同其峰面积与内标峰面积之比呈良好线性关系。天麻与钩藤配伍前后天麻素在肝脏和肾脏的分布有显著性差异。结论:此方法能准确、灵敏地测定大鼠肝、肾组织中天麻素含量。天麻与钩藤的配伍对天麻素在SHR大鼠肝、肾中的分布及归经有显著的影响。 OBJECTIVE: To establish an method for the content determination of gastrodin in liver and kidney tissue of SHR rats and to discuss the meridian distribution of Gastrodia elata by contrasting the differences of gastrodin tissue distribution before and after compatibility of G. elata and Uncariae Ramulus Cum Uncis. METHODS: The contents of gastrodin in liver and kidney tissue of SHR rats were determined by HPLC. The analysis was carried on Phenomenence Luna C18 column(250 min×4.6 mm, 5 μm) with mobile phase consisted of acetonitrile-1% phosphoric acid (3:97) at flow rate of 1.0 mL. min^-1 The detection wavelength was set at 220 nm and column temperature was 30 ℃. The injection volume was 10 μL. RESULTS: The linear rang of gastrodin was 0.312 5-200 μg·mL^-1. The data showed significant difference of the distribution of gastrodin in liver and kidney before and after compatibility of G. elata and Uncariae Ramulus Cure Uncis. CONCLUSION: The method is sensitive and accurate for the content determination of gastrodin in liver and kidney tissue of SHR rats. The compatibility of G. elata and Uncariae Ramulus Cum Uncis shows a significant effect on the meridian distribution of G. elata and the distribution of gastrodin in liver and kidney tissue of SHR rats.
