摘要 目的探讨GDNF在肺腺癌A549细胞株的表达及细胞活力变化.方法体外培养肺腺癌A549细胞株,观察细胞形态变化.取第1代培养后1 d,5 d两个时间点分别计数细胞;MTT检测两时间点细胞活力变化;用免疫组化检测两时间点GDNF免疫阳性细胞数量变化.并进行统计学分析.结果培养5 d组肺腺癌A549细胞株的细胞数量与1 d组相比,明显增加(P<0.05);5 d组的细胞活力较1d组明显下降(P<0.05);GDNF的表达百分数比较显示;5 d组亦较1 d组明显下降(P<0.05).结论体外培养肺腺癌A549细胞株随细胞增殖过程,活力逐渐下降,同时GDNF表达明显下调,提示细胞增殖和细胞活力不一定完全一致,GDNF减少可能与活力减少有关. Objective To study the expression and cell viability changes of GDNF in lung adenocarcinoma cell lines A549.Methods Lung adenocarcinoma cell lines A549 were cultured and morphological changes were observed.Cells were counted and cell viability were detected by MTT method after culturing for 1d and 5 d.timultaneously,GDNF positive cells were detected by immunohistochemistry and statistically analyzed.Results The cell number of lung adenocarcinoma cell lines A549 was significantly increased in 5 d group,compared with1d group(P0.05),but the cell viability in 5 d group was significantly lower than 1d group(P0.05). The positive percentage of GDNF expression in 5 d group was significantly lower than 1 d group(P0.05). Conclusions In the process of cell lines A549 proliferation,cell viability and GDNF expression decrease gradually,suggesting that cell proliferation is not identical with cell viability,the reduction of GDNF may correlate to the decrease of cell vitality.
