摘要
目的研究水溶性黄蘑多糖的提取工艺。方法采用正交设计优化,以多糖得率和多糖含量两项为指标,考察加水量、提取时间、提取温度、提取次数及其交互作用对多糖提取的影响。以半乳糖做标准曲线,测定水溶性黄蘑多糖含量,检测波长490 nm。结果适宜水溶性黄蘑多糖提取的最佳提取工艺条件为:每10 g黄蘑干粉,每次加水150 ml,95℃下提取2 h,提取3次,多糖得率和多糖含量分别为11.6%和8.11%。结论该工艺操作简单,成本较低,适合水溶性黄蘑多糖的提取。
Objective To study the extraction process of the water-soluble polysaccharide in Huangmo.Methods The orthogonal design was adopted to research the effect of water volume,extraction time,extraction temperature,extraction times and their interactions.The standard curve was used to check the content of polysaccharide with galactose as standard sample,and the detection wavelength was 490nm.Results The optimum extraction conditions were as follows: extraction 3 times with water 150ml per 10g Huangmo powder,extraction time 2h,extraction temperature 95℃.The yield rate and content of polysaccharide were 11.6% and 8.11%,respectively.Conclusion The process is suitable for extraction of water-soluble polysaccharide in Huangmo.
