摘要
目的观察黄连解毒汤对自发性高血压大鼠(Spontaneously hypertensive rats,SHR)血管内皮功能的保护作用。方法 24只12周龄雄性SHR随机分为黄连解毒汤组、卡托普利组和模型组3组,8只同龄雄性Wistar-Kyoto(WKY)大鼠为空白对照组,通过BP-6动物无创血压测试仪测定各组治疗前血压,连续给药6周,1次/d,第1,2,4,6周的第7天下午定时测量各组血压,6周后由腹主动脉取血,检测血清血栓素A2(TX-A2)、6酮前列环素(6-K-PG)、一氧化氮(NO)、内皮素-1(ET-1)、超氧化物歧化酶(SOD)、丙二醛(MDA)、血管紧张素Ⅱ(ANG-Ⅱ)和血管性血友病因子(vWF)。结果黄连解毒汤和卡托普利均可降低SHR血压、TX-A2、ET-1、MDA、ANG-Ⅱ和vWF,增加6-K-PG、NO和SOD。结论黄连解毒汤可降低SHR的血压,且对血管内皮功能具有保护作用。
Objective To observe the effects of Huanglianjiedu-Decoction on function of blood vessel endothelium in spontaneous hypertension rats(SHR).Methods Twenty-four SHR were randomly divided into Huanglianjiedu-Decoction-treated group,captopril-treated group and model group;eight Wistar-Kyoto rats were used as blank group.The rats were treated by Huanglianjiedu-Decoction,captopril and distilled water respectively for six weeks.Blood pressure(BP) was measured at the beginning of the experiment and the end of every week in the experiment.After Six weeks later,thromboxane-A2(TX-A2),6-keto-prostaglandin(6-K-PG),nitric oxide(NO),endothelin-1(ET-1),superoxide dismutase(SOD),malondialdehyde(MDA),angiotensin Ⅱ(ANG-Ⅱ) and von willebrand factor(vWF) in plasma were determined respectively.Results Huanglianjiedu-Decoction could decrease BP,TX-A2,ET-1,MDA,ANG-Ⅱ,vWF contents and increase 6-K-PG,NO,SOD levels.Conclusion Huanglianjiedu-Decoction can decrease BP and has a protective effect on blood vessel endothelium in SHR.
