刺五加叶绿体的超微结构与刺五加苷B、E含量的关系 被引量:2

摘要 目的分析不同产地刺五加叶绿体超微结构特征及其与刺五加苷B、E含量的相关性。方法取野生刺五加叶片,利用超薄切片进行透射电镜观察叶绿体超微结构,测量叶绿体长、宽、周长和面积,HPLC法测定刺五加苷B、E含量。结果不同产地刺五加叶绿体超微结构间存在较大差异,叶绿体平均长、宽、周长分别为3.72μm、1.62μm、9.63μm,面积为5.17μm2,叶绿体宽与茎中刺五加苷E含量呈正相关关系,相关系数达0.819 38。结论叶绿体超微结构及其长、宽、周长、面积可以作为鉴定刺五加产地来源的指标,叶绿体宽度可作为茎中刺五加苷B含量的早期鉴定指标。 Objective To analyze the Chloroplast submicroscopic structure of Eleutherococcus senticosus and relevance to eleutheroside B,E among different habitats.Methods Select wild leaves of Eleutherococcus senticosus,and observe Chloroplast submicroscopic structure with ultra-thin slices transmission electron microscope,then measure its length,width,perimeter and the area,analyze eleutheroside B,E content with HPLC method.Results There was much difference among different habitats Chloroplast submicroscopic structure of Eleutherococcus senticosus.The average length,width,perimeter and area were 3.72 μm,1.62 μm,9.63 μm,5.17 μm2,the width of chloroplast was correlated with leutheroside B content in Eleutherococcus senticosus stems,and the correlation coefficient could reach 0.819 38.Conclusion The Chloroplast submicroscopic structure of Eleutherococcus senticosus and its length,width,perimeter and area can be used as indicators to identify the origin of Eleutherococcus senticosus,and the width of Chloroplast can be used as early identification indicator of eleutherosides B content in Eleutherococcus senticosus stems.
