摘要
目的研究刺老苞根皮黄酮类化合物对泼尼松致大鼠骨质疏松的对抗作用。方法将50只大鼠随机分成正常对照组、泼尼松模型组和刺老苞根皮黄酮类化合物高、中、低(10,20,30 mg/kg)剂量组,每组10只。分别灌胃相应药物,1次/d,连续8周后取血清检测生化指标,对骨进行组织切片检查,长度、宽度以及骨质的测量。结果泼尼松模型组大鼠股骨重量、尺骨骨羟脯氨酸和骨钙含量比正常对照组明显降低,胫骨骨髓腔中脂肪组织增多(P<0.01),血浆胆固醇(TG)含量上升、碱性磷酸酶(ALP)和高密度脂蛋白(HDL)含量下降(P<0.01)。刺老苞根皮黄酮类化合物组能有效提高骨的重量和骨质的含量(P<0.01),减少骨髓腔中脂肪的含量,升高碱性磷酸酶和高密度脂蛋白含量(P<0.01)。结论泼尼松可抑制大鼠的骨生长及引起骨丢失,而刺老苞根皮黄酮类化合物可有良好的对抗作用。
ObjectiveTo investigate the effect of Aralia echinocauIis Hand.Mazz flavonoids on osteoporosis induced by prednisone in rats.MethodsFifty SD rats were randomly divided into control group,prednisone group,Aralia echinocauIis Hand.Mazz flavonoids low dosage group(10 mg/kg),middle dosage group(20 mg/kg) and high dosage group(30 mg/kg).Rats in control group were treated with dissolvent,the others were treated with prednisone firstly,and then,the rats in prednisone group were treated with dissolvent,the rats in different Aralia echinocauIis Hand.Mazz flavonoid groups were treated with related dosage extract,respectively,once a day.After 8 weeks,the bone histomorphometric analysis of thighbone was performed,the contents of Ca,P and hydroxyproline(Hyp) in serum were measured,the length and width of thighbone were tested.ResultsThe bone weight,bone Hyp and bone Ca in prednisone group were lower than those in control group(P〈0.01).The fat cholesterol in marrow were increased but alkaline phosphatase(ALP) and high-density-lipoprotein(HDL) were reduced(P〈0.01).Aralia echinocauIis Hand.Mazz flavonoids could increase bone weight,reduce the fat in marrow and increase ALP and HDL(P〈0.01).ConclusionPrednisone can restrain bone growth and increase bone loss,while Aralia echinocauIis Hand.Mazz flavonoids has good protective effect.
