摘要
目的:设计研究一种更适用于霍利斯弧菌快速筛选与检测的选择性鉴别培养基(VhDM)。方法:基于霍利斯弧菌的营养需求、特有的综合酶系统及其代谢特点和对某些抑菌剂的耐受性,进行VhDM的设计与研究,进而对VhDM的显色效果、平板效率、灵敏度和特异性进行评价。结果:VhDM对霍利斯弧菌具有较好的选择性和特异性;培养16~24 h,霍利斯弧菌菌落为红色,其他致病菌菌落呈黄色或不生长;VhDM对霍利斯弧菌的出菌率为(90.65±1.72)%;VhDM的检出限为10 cfu/mL。结论:与常规方法相比,VhDM快速检测霍利斯弧菌的结果易于观察,减少了其他弧菌常伴弧菌的干扰,且降低检测成本、易于操作、减少工作量,具有更高的检测效率和鉴别能力。
Objective: To develop a more economical, simple and rapid separation, identification or detection method of Vibrio hollisae. Methods: We designed and developed the selective differential medium according to the nutritional requirements, the specific enzyme systems, the corresponding metabolisms and the tolerance to different antibacterial ingredients of Vibrio hollisae. We did the tests of color effects, plating efficiency, sensitivity and specificity to evaluate the efficiency of Vibrio hollisae selective differential medium(VhDM). Results: We used VhDM to detect Vibrio hollisae and got good selectivity and specificity. The Vibrio hollisae grown to be red colonies on VhDM after incubation for 16-24 h at 37 ℃, while other species of bacteria no growth or formed yellow colonies. The plating efficiency of Vibrio hollisae on VhDM was (90.65±1.72)% and its detection, limit was 10 cfu/mL. Conclusion: As compared with the conventional methods, the method of rapid separation and detection of Vibrio hollisae with VdDM is simpler operation, lower cost, more convenient for observation, higher sensitivity and specificity. Accordingly, it is suitable for kinds of inspection.
