摘要
目的:通过考察水蛭提取物的抗凝血生物活性,确定水蛭的最佳提取工艺,保证水蛭抗凝血活性成分获得最大保留。方法:选择浸泡时间、煎煮次数、煎煮时间、水与水蛭质量比为考察因素,按L9(34)正交设计因素水平表排列,考察水蛭提取物得率和提取物的凝血酶效价。结果:水蛭的最佳提取工艺为水蛭药材先用10倍量的水浸泡4h,加水煎煮3次,每次煎煮1.5h。结论:水蛭经水提、乙醇精制后凝血酶活性依然保留完好,得率与凝血酶效价呈正相关。此工艺对水蛭提取物的生产简便可行。
Objective: To study the anticoagulant activity of Hirudo extra and determine the optimum extraction of Hirudo in order to ensure the Anticoagulant active ingredients to be greatest retained.Methods:With extraction rate and anticoagulation activity in the extract as assessment indices,the optimum extracting conditions for Hirud were studied by the orthogonal design.Results:The best technology to extract the Hirudo was to use 10 times water to soak the Hirudo for 4 hours,then extract 3 times,each time takes 1.5 hours.Conclusion:Extracted by water and refined by ethanol,the thrombin activity of Hirudo is still retained intact,the yield of thrombin titer is positively correlated.This technology makes the production of leech extraction simple and feasible
