RP-HPLC同时测定黄连解毒汤7种活性成分 被引量:8

Simultaneous Detection of the Contents of Seven Active Components in Huanglian Jiedu Decoction with the Method of RP-HPLC

摘要 目的建立一种同时测定黄连解毒汤中7种活性成分(栀子苷、黄芩苷、盐酸巴马汀、盐酸小檗碱、汉黄芩苷、黄芩素和汉黄芩素)含量的RP-HPLC方法。方法采用迪马Diamonsil C18色谱柱(250×4.6 mm,5μm),以乙腈(A)-0.05%磷酸二氢钾和0.05%三乙醇胺(用磷酸调pH到3.0)(B)为流动相,梯度洗脱:0~9 min,84%B;9~27 min,84%→73%B;27~35 min,73%→70%B;35~39 min,70%→68%B;39~42 min,68%→60%B;42~46 min,60%→52%B;46~53 min,52%→45%B;53~60 min,45%→84%B。流速0.8 ml/min,检测波长254 nm,柱温35℃。结果在60 min内黄连解毒汤中栀子苷、黄芩苷、盐酸巴马汀、盐酸小檗碱、汉黄芩苷、黄芩素和汉黄芩素7种成分被完全分离;回归方程显示7种成分的峰面积与其浓度呈良好的线性相关;7种成分的加样回收率(n=6)为94.92%~106.76%,RSD均小于2.0%。结论利用反相高效液相色谱法,建立了一种简便准确,可同时测定黄连解毒汤中7种活性成分含量的检测方法。 Objective To establish a method of RP-HPLC which can simultaneously detect the contents of seven active components including geniposide,baicalin,palmatine,berberine,wogonoside,baicalein and wogonin in Huanglian Jiedu decoction.Methods The samples were separated by a Diamonsil C18 column(250×4.6 mm,5 μm) with a linear gradient elution under predetermined procedures(0~9 min,84% B;9~27 min,84%→73% B;27~35 min,73%→70% B;35~39 min,70%→68% B;39~42 min,68%→60% B;42~46 min,60%→52% B;46~53 min,52%→45% B;53~60 min,45%→84% B).Additionally,acetonitrile(A) and 0.05% potassium dihydrogen phosphate mixtured with 0.05% triethanolamine(PH was regulated to 3.0 by phosphoric acid)(B)were used as mobile phase at a flow rate of 0.8 mL·min-1.The column temperature was set at 35 ℃.The eluate was detected at the wavelength of 254 nm.Results Seven active components mentioned above in Huanglian Jiedu decoction were completely separated within 60 minutes by the method of RP-HPLC.The regression model suggested a sound linear correlation between the peak areas of seven components and corresponding contents.And the recovery rate(n=6) of the markers listed above ranged from 94.92% to 106.76%.All the relative standard deviations(RSDs) were less than 2.0%.Conclusion The method of RP-HPLC was convenient and accurate to detect the contents of seven active components in Huanglian Jiedu decoction simultaneously.
