摘要
为了探讨H9N2亚型禽流感病毒在MDCK细胞中增殖的最佳条件,将不同毒株、不同接种量的H9N2亚型禽流感病毒接种MDCK细胞单层,接毒后直接加入或吸附1h后加入含有10μg/mL胰蛋白酶的DMEM维持液,每隔24h观察细胞病变,并测定细胞上清液中HA滴度。结果表明,分离株HN04在MDCK细胞中增殖能力较其他2株强,接种后72h可达到6log2;病毒接种细胞的最佳稀释度为103~104之间;病毒接种细胞后吸附与否对病毒滴度无显著差异。
This study aimed to determine the optimal proliferation conditions of H9N2 avian influenza virus in MDCK cells.The proliferation ability of H9N2 avian influenza virus in MDCK cells was evaluated by using different isolates,different inoculation doses,and pre-adsorption for 1 h or non-adsorption in DMEM medium containing 10 μg/mL trypsin.The cytopathic effect(CPE) of MDCK was observed every 24 h,and the HA titer of the supernatant was measured by HA assay.Result showed that the isolate of HN04 showed a little higher HA titer than the other two isolates.The highest HA titer reached 6log2 at 72 h after inoculation.The optimal dilution was 103-104 for all isolates.No significant difference was observed between pre-adsorption for 1 h and non-adsorption.
