摘要
目的观察西维因与氰戊菊酯联合染毒对雄性大鼠睾丸细胞凋亡的影响。方法选择健康成年清洁级雄性SD大鼠40只,按2×2析因设计随机分为4组,分别为阴性对照组(玉米油)、西维因(1/50 LD50,11.2 mg/kg)单独染毒组、氰戊菊酯(1/50 LD50,9.02 mg/kg)单独染毒组和西维因(11.2 mg/kg)+氰戊菊酯(9.02 mg/kg)染毒组,每组10只。采用经口灌胃方式进行染毒,染毒容量为5 ml/kg,每天1次,连续染毒8周。采用Annexin-FITC-PI双标记流式细胞术检测细胞凋亡,区分凋亡与坏死细胞,测定睾丸组织中酸性磷酸酶(ACP)、碱性磷酸酶(AKP)、乳酸脱氢酶(LDH)水平,并观察睾丸组织形态的变化。结果西维因和氰戊菊酯联合染毒对睾丸组织中AKP活力呈现协同作用(P<0.05),对睾丸细胞早期凋亡率呈现拮抗作用(P<0.01),对ACP和LDH活力无交互作用。光镜下可见,西维因和氰戊菊酯联合染毒可引起曲细精管外形不规则、萎缩变性,间质增宽,生精小管管腔内可见各级生精细胞脱落,生精小管生精细胞层数和各级生精细胞数量均减少,细胞排列疏松紊乱,管腔内成熟精子数量减少。结论西维因和氰戊菊酯联合染毒可影响睾丸标志酶活力,促进大鼠睾丸生精细胞凋亡。
Objective To observe the combined effects of carbaryl and fenvalerate on apoptosis in testicular cells. Methods According to 2×2 factorial analysis,forty adult male SD rats were randomly divided into 4 groups,8 in each group, including a control group (given coin oil) and three experimental groups:carbaryl (1/50 LD50,11.2 mg/kg,dissolved in coin oil), fenvalerate (1/50 LD50,9.02 mg/kg,dissolved in coin oil) and carbaryl(11.2 g/kg)+fenvalerate (9.02 g/kg), through gavage, once a day ,for 8 consecutive weeks. The apoptosis of testicular cells in the testis of the rate was detected by Annexin-FITC-PI double labeled cells flow cytometry. The activities of ACP, AKP and LDH were measured in testis homogenate and the histopathological examination of the testis was conducted simultaneously. Results There was antagonism between carbaryl and fenvalerate on the AKP activity and the early apoptosis rate of testicular cells(P〈0.05 ,P〈0.01 ). There was no interaction between carbaryl and fenvalerate on the activity of ACP and LDH in testicle(P〉0.05). Pathological examination showed the somniferous tubules were irregular shape, degenerative atrophy and interstitial substance broadening, shed spermatogenic cells in seminiferoustubules ,the layers and counts of spermatogenic cell reduced and disordered cellular arrangement in testicular tissue. Conclusion Carbaryl combined with fenvalerate can affect the marker enzyme activity,increase the apoptosis rate of germ cellscapable and induce histopathological changes of the testis tissue.
