摘要
目的探讨大鼠血清中Th1/Th2细胞因子表达与供肝非实质细胞(NPC)输注诱导心脏移植免疫耐受的关系。方法建立大鼠心脏移植模型,将40只大鼠随机分成排斥反应组、免疫耐受组,每组20只。观察移植心脏存活时间,并检测受者血清中Th1/Th2细胞因子白细胞介素2(IL-2)、干扰素γ(IFN-γ)、白细胞介素4(IL-4)、白细胞介素6(IL-6)、白细胞介素10(IL-10)的表达水平。结果免疫耐受组供心存活时间为(30.83±2.55)d,排斥反应组为(8.15±2.08)d,差异有显著性(t=24.46,P〈0.01)。术后7、14 d排斥反应组血清Th1细胞因子IL-2、IFN-γ水平均高于免疫耐受组,Th2细胞因子IL-4、IL-10表达水平均低于免疫耐受组,术后7 d血清IL-6表达水平显著高于免疫耐受组(t=4.50~16.20,P〈0.01)。结论免疫耐受的形成与Th1/Th2细胞因子相互作用有关。
Objective To study the relationship between Th1/Th2 cytokine expression in serum and immune tolerance,induced by non-parenchymal cells(NPC) of donated liver,of cardiac allografts in rats. Methods A model of heart transplantation was created in 40 rats,and evenly randomized to rejection group and immune-tolerance group.The survival time of the implanted heart was observed,the expressions of serum Th1/Th2 cytokines: interferon-γ(IFN-γ),interleukin-2(IL-2),interleukin-4(IL-4),interleukin-6(IL-6) and interleukin-10(IL-10),were measured. Results The survival time of heart allografts in tolerance group was(30.83±2.55) d,and that in rejection group was(8.15±2.08) d,the difference being significant between the two groups(t=24.46,P0.01).Seven and 14 days after transplantation,the expression of Th1 cytokines(IL-2,IFN-γ) were much higher in rejection group than in immune-tolerance group,and that of Th2 cytokines(IL-4,IL-10) were lower.Seven days after surgery,the expression of IL-6 in rejection group was much higher than immune-tolerance group(t=4.50-16.20,P0.01).Conclusion The formation of immune tolerance is associated with interaction of Th1/Th2 cytokines.
