摘要
目的探讨胃癌组织中Fas相关因子1(FAF1)蛋白表达与幽门螺杆菌(Hp)感染之间的相关性及其可能的作用机制。方法免疫组化检测40例胃癌手术标本和相应的正常胃黏膜组织中FAF1蛋白的表达,脱氧核糖核酸末端转移酶介导的缺口末端标记(TUNEL)法检查细胞凋亡,HE、甲苯胺蓝染色和Warthin-Starry(W-S)银染法检测Hp感染。结果在40例胃癌组织中FAF1的阳性表达率为37.50%,而在40例正常胃黏膜组织中FAF1阳性表达率为72.50%,二者差异有统计学意义(P<0.05);FAF1蛋白表达与肿瘤浸润深度、大小、临床分期和淋巴结转移无明显相关性(P>0.05),但与胃癌的分化程度有关,低分化胃癌组织FAF1阳性表达率显著低于高、中分化胃癌组织(P=0.010 2)。FAF1表达阴性组的胃癌细胞凋亡指数显著低于FAF1阳性组(P=0.011 0)。Hp阳性胃癌组织的FAF1阳性表达率明显低于Hp阴性者,二者差异有统计学意义(P<0.05)。结论胃癌的发生、发展可能与FAF1的低表达有关,Hp感染可能通过下调FAF1蛋白的表达影响胃癌的分化和凋亡而发挥作用。
Objective To investigate the expression of Fas associated factor 1(FAF1) protein in gastric cancer and the relationship with Helicobacter pylori(Hp) infection,as well as its possible mechanism.Methods Immunohistochemistry was used to detect FAF1 expression in 40 gastric cancer tissues and 40 corresponding gastric normal tissue,and terminal deoxynucleotidyl transferase-mediated nick end labeling(TUNEL) was used to detect cell apoptosis,while Helicobacter pylori infection was detected by HE staining,methylbenzene amine blue staining and Warthin-Starry silver staining(W-S) methods.Results FAF1 positive expression rate was 37.50% in 40 cases of gastric cancer tissues,while 72.50% in 40 cases corresponding gastric normal tissues,significant difference was found between the two groups(P0.05);FAF1 protein has no significant correlation in gastric cancer patients by tumor invasion,size,clinical stage and lymph node metastasis(P0.05),but the degree of tumor differentiation.FAF1 expression of low or non-differentiated gastric cancer was significantly lower than that of well-differentiated ones(73.08% vs 64.29%,P=0.010 2).Apoptosis index was significantly lower in FAF1 negative expression group than that in FAF1 positive expression one(P=0.011 0).FAF1 positive expression rate in Hp-positive group was significantly lower than that in Hp-negative group in gastric tissue,and there was a significant difference between the two groups(P0.05).Conclusion Gastric carcinogenesis may be associated with low expression of FAF1,and Hp infection may contribute to the gastric carcinoma differentiation and apoptosis by down-regulating the expression of FAF1 protein.
