摘要
目的:观察不同温度对小鼠胚胎玻璃化冷冻的影响,探索小鼠胚胎玻璃化冷冻的最适宜温度。方法:选用8周龄昆明系小鼠超排卵,取桑葚胚或早期囊胚在不同温度下玻璃化冷冻,解冻后体外培养,观察其48小时发育率和孵化率。结果:25℃时胚胎冷冻效果要优于4℃(P<0.01)及37℃时(P<0.05)。25℃时和对照组之间无差异。结论:温度对小鼠胚胎玻璃化冷冻保存有影响,25℃为最适温度。
[Objective]By an observation of the development of the thawed mouse embryos,to find the most suitable temperature of vitrification.[Method]Mouse monilae or blastocyst from in vivo were frozen by vitrification at 4℃,25℃and 37℃respectively,then the developmental ability of the thawed mouse embryos was assessed.[Result]the hatch rate of thawed mouse embryos vitrified at 25℃is higher than that at 4℃or 37℃.[Conclusion]Temperature is ah important element for the vitrification of mouse embryo,and the 25℃is the most suitable temperature.
