摘要
应用激光扫描共聚焦显微镜对一串红花粉粒进行连续“光学切片”扫描,利用Laser-Sharp图像处理软件重建其三维立体空间结构。结果显示,重建的图像为高分辨率的三维原色图像,并可沿X、Y、Z轴旋转或任意角度显示。叙述了激光扫描共聚焦显微镜成像原理、特点以及在植物孢粉学研究中的应用前景。
Laser scanning confocal microscopy was used to acquire sequential 'optical sections' of the pollen of Salvia splendens Ker-Gawl. The three-dimensional structure of the pollen was then reconstructed by means of Laser Sharp software. The result demonstrated that the reconstructed 3-dimensional images were of primary color images with high resolution power, and could be rotated and observed from any angle in the 3D space. The optical principle and characteristics of LSCM with the prospect in applications of LSCM in palynology research were also described in the paper.
