摘要
绿豆种子于1994年经返回式卫星搭载后,经过三年的地面种植和筛选得到了基本稳定的长荚型突变系,该突变系平均荚长16cm左右,每荚种子粒数在15—19粒.对突变系和其原始对照品系进行了RAPD(RandomlyAmplifiedPolymorphicDNA)分析.共用了100个10-merOperon引物,其中三个引物在突变系和原始品系之间扩增出了多态性产物.所分析的三个突变系形态一致,彼此之间没有发现明显差异,但它们与原始对照之间都有着共同的差异,这表明突变品系已基本稳定.其中两个多态性产物的克隆工作已完成,另外一个扩增产物的克隆工作正在进行中.
Mung beau seeds were carried into space in 1994 by a recoverable Satellite.After three years groud cultivation and selection, a long pod mutant line was oaftamed. The pod length of this mutant line ranged from 13 to 17cm long, in the average of 16cm. The seed setting number in each pod ranged from 15 to 19, in the average of 17. RAPD (Randomly Amplified Polymorphic DNA) anajysis wasperformed between this mutant line and its original CK line. Total of 100 10-inerOperon primers were screened, of them, 3 amplified polymorphic products. Three samples with same phenotype and about the same pod length selected from this mutant line showed no polymorphism in their RAPD pattern. But they differ from the original CK line with same polymorphism. These results indicated that the long-pod mutant line may be close to homologous. Two of the three polymorphic products were cloned already. Cloning work of the other polymorphic products is being performed.
