摘要
目的:通过测定慢性阻塞性肺疾病(COPD)患者肺组织中水通道蛋白5(AQP)、钠泵α1亚单位蛋白的表达及与肺功能变化的关系,明确AQP5、钠泵α1亚单位在COPD发生发展中的作用。方法:选取肺叶切除术患者肺组织标本52例,其中,对照组25例,COPD组27例;免疫组化法检测AQP5、钠泵α1亚单位蛋白表达。结果:与对照组相比,AQP5蛋白在COPD组表达降低(P<0.05),在COPD中度组较轻度组表达亦降低,两组比较,差异有统计学意义(P<0.05)。AQP5表达改变与FEV1/FVC、FEV1预计值呈正相关关系(Rs=0.717,P<0.05;Rs=0.500,P<0.01)。钠泵α1亚单位在COPD组患者的表达与对照组相比差异无统计学意义(P>0.05)。结论:AQP5参与了COPD患者气道黏液分泌的病理过程并与COPD气流受限有关,钠泵α1亚单位在COPD的发病机制中可能起辅助作用。
Objective: To study the role of AQP5 and sodium pump α1 subunit in development of COPD by measuring AQP5 and sodium pump α1 subunit protein expression and their relationships with pulmonary function changes in COPD patients.Methods: The sample from 52 cases of pulmonary lobectomy patients,including 25 cases of control group and 27 cases of COPD group,each sample was stained by immunohistochemical staining to detect AQP5 and sodium pump α1 subunit protein expression.Results: Compared with the control group,AQP5 protein expression decreased in the COPD group(P0.05);moreover,compared with mild group,AQP5 also decreased in moderate COPD group(P0.05).Change of AQP5 expression was positively correlated with predicted values of FEV1/FVC and FEV1(Rs=0.717,P0.05;Rs=0.500,P0.01);sodium pump α1 subunit expression in COPD patients showed no significant difference compared with the control group(P0.05).Conclusion: AQP5 participates in pathological process of airway mucus secretion in COPD patients and has the relationgship with airway obstruction of COPD;sodium pump α1 subunit may play a minor role in the pathogenesis of COPD.
