摘要
应用改良的Martin(1993)定量反转录-聚合酶链式反应(reversetranscription-poly-merasechainreaction,RT-PCR)方法,测定了大鼠ETA型受体((Endothelin-Areceptor,ETA-R)mRNA在心血管系统细胞中的分布。结果证明,ETA-RmRNA广泛地分布在心血管系统细胞中.其中以平滑肌细胞中含量最高。内皮素及其受体的配基可以促进血管平滑肌细胞(vascularsmoothMusclecell,VSMC)中ETA-RmRNA的表达。此外,还发现在自发性高血压动物的血管平滑肌细胞中ETA-R基因的高表达可能是高血压血管高反应性的一个重要机制。

Reverse Transcription- Polymerase Chain Reaction (RT-PCR ),expression
of endothelin- Areceptor(ETA - R) mRNA in the cells of rat
cardiovascular system was studied.It was found that(ETA- R) mRNA is
widely distributed in the cells of rat cardiovascular system.Among
them,(ETA- R)mRNA level is the highest in the smooth muscle
cells(SMCs) .Endothlin can significantly increase the(ETA - R) mRNA
level in SMC,and in addition,(ETA- R)mRNA levels are
significantly increased in spontaneously hypertensive rat VSMC,These
data show that the high expression of( ETA - R) gene in SMC may play
an important role in the pathogenesis of hypertension.
