人胃癌Ras基因多态性分析

摘要 用C-Ha-ras癌基因作探针,研究各型胃癌DNA对限制性核酸内切酶BamHI和SacI的多态性。结果发现,在各型胃癌ras基因,出现5种BamHI片段和4种SacI片段,这些等位片段似乎与胃癌的病理组织学分型无明显关系。另外,Ha-ras基因BamHI片段及SacI片段在胃癌组织及癌旁“正常”胃粘膜的出现规律基本一致。提示;用癌旁胃粘膜作为健康对照组是不足取的。 RFLP was studieds in tumor DNAs of 18 ratients with gastric cancer.Five alleles(BamH I Digestion) and four alleles (Sac I Digestion) were foundwhen c-ha-ras oncogene probe was used. The results showed that no strikingrelationship existed between RFLP and the histopathological pattern of thecarcinomas. In addition, the RFLP of DNAs in the cancer tissue was almostthe same as that in the 'normal mucosa' beside cancer tissue. It suggested thatusing' normal mucosa'beside cancer tissue as control was unbelieveable.
