摘要 目的探讨低强度脉冲超声(LIPU)对体外分离培养的骨髓间充质干细胞增殖效应(BMSC)的影响。方法将体外分离培养的第3代BMSC随机分为2组:LIPU辐照组和对照组。每天镜下观察细胞生长情况;四甲基偶氮唑蓝法检测细胞增殖活性;流式细胞仪检测各组细胞周期及增殖指数。结果 LIPU能够明显促进BMSC增殖,LIPU组辐照后细胞分裂增殖活性与对照组比较明显活跃,细胞集落更为密集;MTT法检测LIPU组于辐照前、辐照后1,3,5,7 d的细胞增殖活性均明显高于对照组(P<0.05);流式细胞仪检测提示LIPU组辐照后细胞增殖指数明显高于对照组(P<0.05)。结论低强度脉冲超声波对骨髓间充质干细胞的增殖具有明显的促进作用。 Objective To investigate the effect of bone mesenchymal stem cells (BMSC) proliferation by low intensity pulsed ultrasound (LIPU) in vitro. Methods The third generation of BMSC was randomly divided into 2 groups:LIPU groups and control groups. Growth of cells was observed daily under light microscope and the cell proliferation was determined by MTT;Cell cycle changes and proliferation index (PI) were detected using the flow cytometry. Results The LIPU could promote the proliferation of BMSC. Cell proliferation of the LIPU group was significantly more active than that of the control group and more intensive cell colonies formed in LIPU groups;MTT test showed that cell proliferation in the LIPU group of 1,3,5,7 d post-irradiation was significantly higher than that in of the control group (P 〈 0.05) ; Cell cycle changes tested by the flow cytometry showed that the cell proliferation index of LIPU groups was significandy higher than that of control groups after irradiation (P 〈 0.05 ). Conclusion The LIPU could promote the proliferation of BMSC.
