摘要
目的观察SKI-Ⅱ对胃癌SGC7901细胞内血管内皮生长因子(vascular endothelial growth factor,VEGF)表达的影响,并对其机制进行初步研究。方法常规培养胃癌SGC7901细胞,给予不同浓度SKI-Ⅱ及顺铂(DDP)干预,MTT法测定各组胃癌SGC7901细胞增殖情况,免疫组化和Western blot检测药物对SphK1、VEGF表达的影响。结果 MTT检测显示SGC7901细胞与不同浓度的SKI-Ⅱ共同培养,细胞的生长受到不同程度的抑制,其作用具有剂量依赖性,并随时间的延长逐渐增强,免疫组化及Western blot检测显示SKI-Ⅱ可显著下调肿瘤细胞SphK1和VEGF蛋白的表达,单用DDP对SphK1、VEGF无作用。结论 SKI-Ⅱ可以通过下调VEGF和SphK1蛋白表达而有效抑制胃癌SGC7901细胞增殖。
Objective To investigate the effects and mechanism of SKⅠ-Ⅱ on the expression of vascular endo- thelial growth factor (VEGF) in gastric cancer SGC -7901 cells. Methods SGC7901 gastric cancer cells in vitro were treated with different concentrations of SKⅠ-Ⅱ or DDP. Cell proliferation was analyzed by MTT assay. And SphK1 and VEGF protein expression was assessed by immunohistochemistry and western blot. Results SGC7901 cell proliferation was inhibited by SKⅠ-Ⅱ in dose - dependent and time - dependent manners. The expression of SphK1 and VEGF in SGC7901 cells was reduced by SKⅠ-Ⅱ , but not by DDP. Conclusion SKⅠ-Ⅱ down - regulates SphK1 and VEGF proteins, thus inhibits the proliferation of SGC7901 cells.
