摘要
[目的]从土壤链霉菌CaiF1发酵液中分离纯化抗菌物质,并研究该物质的活性。[方法]采用乙酸乙酯萃取、大孔吸附树脂吸附、硅胶层析和制备型高效液相色谱(HPLC)等分离技术纯化抗菌物质,以金黄色葡萄球菌、白粉病菌为指示菌进行活性研究。[结果]土壤链霉菌CaiF1发酵液中抗菌物质得到分离纯化;该物质在pH 2.0~10.0、100℃和紫外照射24 h条件下抑菌活性几乎不变,对白粉病的防治效果达69.7%。[结论]分离纯化的土壤链霉菌CaiF1抗菌物质具有很好的稳定性,为其结构鉴定和开发应用提供理论依据。
[ Objective ] Purification and activity of antibacterial substances derived from soil Streptomyces sp. CaiFl were studied. [ Method ] The antibacterial substances were separated and purified by Ethyl acetate extraction, macroporous adsorptive resin, silica gel chromatography and preparative high performance liquid chromatography ( HPLC ), taking Staphylococcus aureus and Powdery mildew as activity indicating bac- terial. [ Result] Antibacterial substances were purified and the stability analysis of the extracts form Streptomyccs CaiF1 fermentation broth showed stable at pH 2.0 - 10.0, 100 ~C and changed very little under UV treatment for 24 h. Inhibition rate of powdery mildew was 98.37%. [ Conclusion] The purified antibacterial substances showed good stability, which provided theoretical foundation for their structural identifica- tions and future applications.
