摘要
目的建立HPLC法同时测定野菊花中蒙花苷和木犀草素的含量。方法采用RP-HPLC法,对样品处理方法、色谱柱和流动相等影响分离效果的主要因素进行了优化。选用Thermo BDS HYPERSILC18色谱柱;乙腈-0.5%冰乙酸溶液(32∶68)为流动相;检测波长348nm。结果蒙花苷的线性范围0.24~1.22μg,r=0.99995;平均回收率98.15%,RSD为1.42%(n=6);木犀草素的线性范围0.012~0.085μg,r=0.99996;平均回收率98.73%,RSD为1.02%(n=6)。结论该方法简便、实用、结果可靠,分离效果好。
Objective To establish a method for determination of linarin and luteolin in Flos Chrysanthemi Indici by HPLC.Methods The main factors of influencing chromatographic separation effects(extracting method and chromatographic conditions) were optimized.Thermo Hypersil BDS C18 column was used with a mobile phase composed of acetonitrile-0.5% glacial acetic acid(32∶68).The detection wavelength was at 348nm.Results The calibration curve of linarin was linear in 0.24~1.22μg,r=0.99995.The average recovery was 98.15%,RSD=1.42%(n=6).The calibration curve of luteolin was linear in 0.012~0.085μg,r=0.99996.The average recovery was 98.73%,RSD=1.02%(n=6).Conclusion This method is simple and accurate,and the results were stable with a good repeatability.It can be well applied for the quality control of Flos Chrysanthemi Indici.
