摘要
目的:观察水蛭素对H22荷瘤小鼠的抑瘤作用,并探讨其作用机制。方法:接种H22肝癌细胞于小鼠右腋下,形成实体瘤模型。将40只小鼠随机抽样分为4组,盐水对照组(A组)、氟尿嘧啶(5-Fu)组(B组)、水蛭素高剂量组(C组)、水蛭素低剂量组(D组),每组10只。连续用药14 d,次日处死取瘤称重,计算抑瘤率,免疫组化技术检测Histone H1的表达。结果:B、C组对小鼠肝癌细胞的生长抑制率分别为52.4%、37.98%。Histone H1在B、C、D组中均呈阳性表达,与对照组比较差异有统计学意义(P<0.01)。结论:水蛭素对肝癌细胞有抑制作用,其作用机制可能与影响Histone H1参与调控基因表达有关。
OBJECTIVE: To observe the antitumor effect of hirudin in mice bearing H22 tumor cell and explore its mechanism.METHODS: The right armpits of 40 mice were injected with H22 hepatoma cells to establish solid tumor model,then the mice were randomized into 4 groups of 10 each: saline control group(Group A),5-Fu group(Group B),high dose hirudin group(Group C),and low dose hirudin group(Group D).After drug exposure for 14 days,the mice were put to death on the following day for sampling and weighing of the tumor,then the tumor inhibition rate was calculated and the expression of Histone H1 was detected using immunohistochemistry technique.RESULTS: The tumor inhibition ratio in Group B and Group C were 52.4% and 37.98%,respectively.There were positive expressions of Histone H1 in Group B,C and D,all showing significant differences as compared with the control group(P0.01).CONCLUSION: The inhibitory effects of hirudin on hepatoma cells is possibly due to its impact on Histone H1 that is essential for the regulation of gene expression.
