活血解毒方对糖尿病大鼠视网膜PEDF表达的影响 被引量:8

Effect of Huoxue Jiedu Formula on Expression of Pigment Epithelium Derived Factor in Retina of Rats with Diabees

摘要 目的:观察活血解毒方对糖尿病大鼠视网膜色素上皮衍生因子(PEDF)的影响。方法:采用链脲佐菌素(65 mg.kg-1)一次性ip SD大鼠建立糖尿病大鼠模型,随机分为模型组和活血解毒方高、低剂量组(15.8,7.7 g.kg-1)及导升明组(0.167 g.kg-1),另设正常对照组。ig给药,20周后处死动物。腹主动脉取血检测血清中PEDF;摘除眼球应用免疫组织化学观察PEDF蛋白的表达;取视网膜组织,采用荧光定量PCR法检测视网膜PEDFmRNA的表达。结果:糖尿病模型组大鼠血清中PEDF的含量(89.96±18.12)ng.L-1,比正常组降低(P<0.01),与模型组相比,活血解毒方各剂量组的含量均上升。糖尿病模型组大鼠视网膜LSAB法标记染色病理切片中PEDF表达降低,积分吸光度(IA)为(10.49±2.37),与正常组相比(41.75±10.25)有显著性差异(P<0.01),活血解毒方各剂量组与模型组相比显著升高(P<0.01)。与正常组相比,PEDFmRNA在糖尿病模型组的表达降低,而活血解毒方各剂量组的表达比模型组升高。结论:活血解毒方能够升高糖尿病大鼠视网膜PEDF蛋白和mRNA的表达,从而对糖尿病大鼠视网膜起保护作用。 Objective:To observe the effect of Huoxue Jiedu formula on expression of pigment epithelium derived factor(PEDF)in retina of rats with diabetes.Method: A single intraperitoneal injection of streptozotocin(65 mg·kg-1)was used to induce diabetic rats.Model rats were randomly divided into diabetes mellitus(DM)group,Huoxue Jiedu formula groups(15.8,7.7 g·kg-1) and Doxium group(0.167 g·kg-1).The normal group and model group were given normal water by intragastric administrate.All rats were killed after 20 weeks.The serum PEDF levels were measured.Immunological histochemistry assay was used to observe the expression of PEDF.The expressive abundance of PEDFmRNA in rats' retina was detected by real-time quantitative PCR.Result: The level of PEDF in serum,the retinal PEDF protein expression and PEDFmRNA were significantly lower in DM rats than in control group(P0.01).The level of PEDF in serum of Huoxue Jiedu formula groups were higher than those in model group.Compared with the model group,the expression of PEDF in Huoxue Jiedu formula groups increased evidently(P0.01)and the expression of PEDFmRNA were added than those in model groups(P0.01).Conclusion: Huoxue Jiedu formula medicine can increase the expression of PEDF in retina of rats with diabetes.
