摘要
目的:研究余甘子不同提取部位提取物对大鼠急性痛风性关节炎的作用。方法:实验分为空白对照、模型、秋水仙碱和余甘子醇提物、石油醚、乙酸乙酯、正丁醇、水相部位组,采用尿酸钠诱导大鼠痛风性关节炎模型,测定大鼠关节肿胀度、观察大鼠步态、测定关节组织前列腺素E(2PGE2)和血清肿瘤坏死因子-α(TNF-α)含量,考察余甘子不同部位提取物对痛风性关节炎的作用。结果:与模型组比较,余甘子醇提物、乙酸乙酯部位、正丁醇部位、水相部位的踝关节肿胀度显著降低(P<0.05);各提取物组步态评分显著降低(P<0.05);醇提物、乙酸乙酯部位、石油醚部位、正丁醇部位的PGE2含量显著降低(P<0.01或P<0.05);正丁醇部位、乙酸乙酯部位、石油醚部位组大鼠血清的TNF-α含量显著降低(P<0.05)。结论:余甘子提取物正丁醇和乙酸乙酯部位对大鼠痛风性关节炎有明显的抗炎镇痛作用,其作用机制是通过抑制局部组织和血清炎症介质(PGE2和TNF-α)的释放而减轻炎症反应。
OBJECTIVE: To study the effects of different extract parts from Phyllanthus emblica on acute gouty arthritis in rats. METHODS: Rats were divided into blank control group, model group, colchicine group and methanol extracts group, petroleum ether part group, ethyl acetate part group, n-butyl alcohol part group, water-phase part group of P embilica. Gouty arthritis model of rat was induced by sodium urate in above groups. The swelling degree of joint, gait, the contents of PEG2 and serum TNF-α were observed and determined. The effects of different extract parts from P emblica on gouty arthritis were investigated. RE- SULTS: Compared with model group, the swelling degree of ankle joint of methanol extracts of P. emblica group, ethyl acetate part group, n-butyl alcohol part group and water-phase part group decreased significantly(P〈0.05).Gain score of each extracts group decreased significantly(P〈0.05).PEG2 content of methanol extracts of P emblica group, ethyl acetate part group, petroleum benzine group, n-butyl alcohol part group decreased significantly(P〈0.05).TNF-α content of acetic ether group, n-butanol group, petro- leum benzine group also decreased significantly(P〈0.05). CONCLUSION: Ethyl acetate part and n-butyl alcohol part of the extracts of P emblica show obvious anti-inflammatory and analgesic effect on gouty arthritis in rats. The mechanism of relieving inflammation reaction is to inhibit the release of PGE2 and TNF-α in local tissue and serum.
