摘要
目的探讨成人隐匿性自身免疫糖尿病与调节性T细胞(Treg)及相关基因Foxp3表达关系。方法采集LADA组、T1DM组、T2DM组和正常对照组新鲜外周血,密度梯度分离单个核细胞,流式细胞术检测4组CD3、CD4、CD25 T淋巴细胞亚群数量,计算Treg数量,RT-PCR检测转录因子Foxp3 mRNA的表达水平。结果 LADA组和T1DM组外周血CD4+T细胞明显高于T2DM组和正常对照组(P<0.05)。LADA组外周血Treg细胞高于T1DM组(P>0.05),明显低于T2DM组和正常对照组(P<0.05)。T1DM组外周血Treg细胞明显低于T2DM组和正常对照组(P<0.01),T2DM组和正常对照组之间无统计学差异(P>0.05)。LADA组Foxp3 mRNA明显高于T1DM组(P<0.05),明显低于T2DM组和正常对照组(P<0.05)。T1DM组Foxp3 mRNA明显低于T2DM组和正常对照组(P<0.01),T2DM组和正常对照组之间无明显统计学差异(P<0.05)。结论 LADA患者Treg细胞数量和活性降低,可能是其发病机制之一。
This study aim to investigate the relationships of Latent autoimmune diabetes in adults(LADA) with regulatory T cells(Treg) and expression of its related gene Foxp3.Peripheral blood samples were collected from LADA group,T1DM group,T2DM group and normal control group,for getting mononuclear cells by density gradient separation.The numbers of CD3,CD4,CD25 T lymphocyte subpopulations of the 4 groups were detected with flow cytometry and the number of Treg was calculated.The expression level of transcription factors of Foxp3 mRNA were detected with RT-PCR.Results demonstrated that the number of peripheral blood CD4+T cells of LADA group or T1DM group was significantly higher than that of T2DM group or normal control group(P 〈 0.05);The number of Treg of LADA group was higher than that of T1DM group(P 〉 0.05),but significantly lower than those of other groups(P 〈 0.05);the number of Treg of T1DM group was significantly lower than that of T2DM group or normal control group(P 〈 0.01).There was no difference of Treg number of had between T2DM group and control group(P 〉 0.05).The expression level of Foxp3 mRNA of LADA group was significantly higher than that of T1DM group(P 〈 0.05),but significantly lower than those of other groups(P 〈 0.05).The expression level of Foxp3 mRNA of T1DM group was significantly lower than that of T2DM group or control group(P 〈 0.01).The expression level of Foxp3 mRNA in T2DM group and control group showed no significant statistical difference(P 〉 0.05).All these results indicate that the reduction of the amount and activity of CD4+CD25+regulatory T cells maybe one of the pathogenesis of LADA.
