摘要
以衡水湖湖底污泥为初始样品,经富集、分离纯化、尼罗蓝染色,远红外光谱及气相定性、定量分析筛选出具有PHA合成能力的菌株,结果表明,在分离得到的菌株中有5株菌具有PHA合成能力,其编号分别定为SJ-2、SJ-5、SJ-9、SJ-11、SJ-20,5株菌产PHA的远红外扫描图谱相近,均在1740cm-1处有较强吸收峰,此处为PHA结构的特征峰。5株产PHA的量不同,SJ-9菌产量最高为1.85g/L,菌株SJ-9的最适生长环境为温度为28℃、pH值8、转速150r/min、装液量100mL/500mL。
Five PHA-producing microbial strains, named S J-2, SJ-5, S J-9, S J-11 and SJ-20, were isolated from the sludge sampled from the bottom of Hengshui Lake(China)by enrichment, purification, Nile blue staining, and infrared spectrocopy and gas chromatography analysis. The IR spectra of the 5 strains were similar and had a characteristic sharp PHA absorption band around 1740cm-1. The PHA synthesis abilities of the 5 strains were dif- ferent. In this study, the maximum PHA production was obtained from SJ-9 which showed 1.85g/L cultivated at 28℃, pH value 8 and 150r/min in 500ml Eflenmeyer flask containing 100ml medium.
